Ribonuclease T1 is an endonuclease that specifically degrades single-stranded RNA at the G residue site. This enzyme cleaves the phosphodiester bond between the 3 ′-guanosine residue and the adjacent nucleoside 5 ′-OH group by forming a nucleoside 2 ′,3 ′-cyclic phosphate intermediate. The reaction product is a 3 ′-GMP terminal oligonucleotide. RNase T1 exerts activity without the participation of metal ions.
